Cytomixis in maize haploids
WebMaize doubled haploid (DH) technology provides fixed, pure lines from a donor parent. Protocols for breeding of DH lines are available for over 250 crop species, and over 300 DH-derived cultivars have been developed in 12 species worldwide (Forster and Thomas, 2005). WebFour of them are utilized to produce maize haploids, the first of which involves obtaining haploids by tissue culture from megasporocytes and microsporocytes of maize (Barloy 1989; Petolino et al. 1992). Ao et al. culture the maize unpollinated ovaries on MS and N6 medium and obtain nearly 2% haploid plants (Ao et al. 1982).

WebOct 23, 2024 · Haploid inducer lines are routinely used in plant breeding for maize only, and thus represent an exception. Maize haploid inducer lines all derive from a particular genotype discovered in the 1950s that possesses the ability to induce the development of haploid embryos on a maize line of interest upon pollination with the inducer pollen. WebOct 15, 2024 · Several hypotheses on the possible mechanisms of haploid maize development via pollination with haploinductors have been proposed. Some researchers believe this phenomenon to be connected with sperm cell function disorders, such as incapacity to movement or convergence and fusion with ovule [ 10 ]. WebThe induced haploids are doubled by colchicine and immediately tested for agronomic performance. The traditional technology is based on an inbred called Stock 6 that induces haploids when crossed as a male (1) and, in modern lines, has been reported to induce haploids at frequencies as high as ~15% (2). The key underlying gene, called

WebHaploid induction and chromosome doubling of induced haploids are the two main steps of doubled haploid creation. Both steps have their own complexities. Chromosome doubling of induced haploids may happen spontaneously, although usually at a low rate.
